Dick Advocaat's side – who are goal difference ahead of second-placed FC Twente – are likely to lose their lead in the league at the weekend after suffering a surprise defeat at the Philips Stadion in Eindhoven.
PEC Zwolle – playing in the top-flight for the first time since 2004 this season – had not won in their past five Eredivisie matches but started brightly.
And they look the lead after 25 minutes through Fred Benson, although PSV levelled just before half-time.
Slovenian striker Tim Matavz was the scorer, converting Dries Mertens' assist.
It was expected PSV would go on and win but Benson had other ideas, netting his second goal nine minutes into the second half.
Wiljan Pluim got in on the act shortly after and doubled PEC Zwolle's lead before a red card to PSV defender Erik Pieters made things even tougher for the hosts.
And there was to be no fairytale story this time for PSV as the visitors held on for an important three points that lifted them up to 13th on the table.
